Breakthrough Triple Therapy Cures Pancreatic Cancer in Mice Without Major Side Effects

In a landmark preclinical study, researchers have achieved a complete and lasting eradication of pancreatic cancer tumours in mice using a novel triple combination therapy, offering fresh hope in the fight against one of the deadliest forms of cancer.

Published in the Proceedings of the National Academy of Sciences (PNAS), the research was led by Dr Mariano Barbacid and his team at the Spanish National Cancer Research Centre (CNIO). The scientists tackled a major obstacle in pancreatic cancer treatment, treatment resistance, by targeting the cancer's molecular mechanisms at multiple levels.

A New Strategy Against Resistance

Existing targeted drugs can initially shrink pancreatic tumours, but often stop working as the cancer adapts. In contrast, the CNIO team developed a therapy that simultaneously blocks the cancer-promoting KRAS signalling pathway at three points, significantly reducing the tumour's ability to evade treatment.

The triple therapy combines:

  • An experimental KRAS inhibitor (daraxonrasib),
  • An existing lung cancer drug (afatinib), and
  • A protein degrader (SD36).

When given together to three different mouse models of pancreatic ductal adenocarcinoma (PDAC), the most common and lethal type of pancreatic cancer, the treatment caused dramatic tumour regression with no significant toxic side effects.

Dr Barbacid and colleagues observed that the tumours not only shrank but disappeared, and most importantly, did not develop resistance to the treatment, a persistent issue with current therapies. The study's authors describe the results as a foundation for designing combination therapies that could improve survival in humans.

What This Means for Patients

While the findings represent a major scientific milestone, the researchers note that human clinical trials have not yet begun. Optimising triple therapy for safety and effectiveness in people will take time, and regulatory approval remains a long way off. However, these results pave the way for next-generation treatment strategies that could significantly improve patient outcomes.
